Synopsis
Yanagisawa wakes up, and realizes that Kaito has been defeated. Kurama tells him they've used the keys from him and Kaito to open two of the three doors, but they found a note on the third door stating that if it's opened without Yana's permission, Yusuke dies. Yana lets them through, and they see several staircases. Yana explains that Yusuke is on the second floor, but they will each have to take a seperate staircase to get to him. Botan gives everyone some marking seals to apply to each other -they write their names on each one, and Botan explains that they also show the health status of a person by changing colors. Hiei is reluctant to have any, until Kurama applies one to him before he can stop him.
The group start using the stairs, which have strange effects and other illusions. They eventually make their way to the room where Yusuke and Kido are, but Kido reveals that one of the four is an imposter. Yana's ability, copy, allows him to perfectly copy someone. Kido tells Yusuke to figure out who the imposter is, and gives him 10 minutes to ask his friends questions so he can try and figure it out. If he doesn't do it, he'll have the person they captured killed. Yusuke starts asking questions, but nothing he asks really helps (he asks Kuwabara what his birthday and blood type are, only to realize he himself doesn't know anyway) or gets angry responses (he tries asking Botan what her three sizes are).
He asks Hiei what his sisters name is, but Hiei says he doesn't have to say, as Yusuke already knows. Yusuke knows that's the kind of response the real Hiei would say, but Kuwabara is more concerned at the idea that Hiei has a sister. He becomes more distraught when he finds out seemingly everyone but him knows who Hiei's sister is. Yusuke then notices the seals, and realizes that only the person that applied them can take them off. The group try it, but they're all able to remove the seals they applied. Kido says that time's up, and tells Yusuke to punch the person he thinks is the imposter as hard as he can -it's the only way to beat Yana. Yusuke charges forward...
...and hits "Kuwabara", who changes into Yana. Kido asks how he knew, and Yusuke says that he thought Kurama and Hiei wouldn't be stupid enough to let themselves be captured (Kurama asks Hiei if his ears are burning), whilst if they'd captured and harmed Botan, the weakest of the group, he'd really have had to come back and crush them. He thought it was Kuwabara because he was the easiest to hit -he's hit him a lot in the past, after all. The group are about to turn on Kido, but he says the real Kuwabara will soon be returned. But first, he has the person who was behind all of this come out: Genkai! Kurama thought she might have been behind this, as their opponents could have defeated them very easily in a number of ways. But since they didn't, it must have meant that there was some purpose for this. And since Genkai knows them all so well, he couldn't think of anyone else who would do this in such a roundabout way. Genkai tells them that whilst they're strong, there are still people out there who have means to kill them.
Kido explains to them that he and his friends only recently discovered their powers, and had gone to Genkai seeking help. Genkai says the cause of these powers is a hole to the Demon Realm. Kuwabara comes back and says Genkai told him everything -someone is trying to open a doorway to the Demon Realm. As he goes to get his clothes back from Yana, Botan asks who it could be, as Sakyo was meant to have died. Genkai says Sakyo died for sure, so this means that there's someone out there who wants to carry out Sakyo's wishes. Yusuke wonders who this person could be. Who is this person who is carrying on Sakyo's wishes?
-The scene in the anime where Kido steps out of Yusuke's shadow, and nearly gets punched for it, is not in the manga.
-In the anime, the sequence where Botan explains what the marking seals do is more elborate, with her trying one on Kuwabara, and then there are some funny images of Kuwabara in various states as she says about the health condition colors. In the manga, she just says these things without the visual aids.
-In the manga, Kurama's seals are on his arm, in the anime they're on his chest.
-The scenes showing everyone using the different stairs isn't in the manga.
-In the manga, Yusuke asks an additional question to Kuwabara: what's his sister's name? Kuwabara correctly says it's Shizuru. This is not in the anime.
-The manga does not show what Kuwabara thinks Hiei's sister could look like, but the anime does.
-In the manga, Yana gets back up after being punched, and stands next to Kido as Genkai explains things. In the anime, he stays down.
-In the manga, Kuwabara is wearing his underwear, t-shirt and socks when he comes back. In the anime, he's only in his undies.
-Kuwabara also lightly hits Yana to get his clothes back in the manga, in the anime he doesn't hit him.
